Distinguish Virgin Drosophila

In fly genetics or fly behavior study, we often need to sort virgin females. Here, I will demo how to identify the virgins with simply two steps!
Step one: find the dark spot in the abdomen
- Get the newly emerged adult flies, preferably less than 6h after eclosion. (For example, when there is a peak of dark black color pupae, start sorting every 6h during the day. Remember to dump all non-virgin flies)
- Anesthetize flies with CO2, use a paintbrush to check flies’ abdomen under a microscope. If there is black spot or “meconium” , then the flies are considered as newly emerged and unmated flies because meconium is the remains of their last meal before pupation. The figure below shows a male’s meconium (left) and a female’s meconium (right) of Drosophila suzukii.
Step two: distinguish females from males
Continue to use D. suzukii (Spotted Wing Drosophila) as an example. After sexual maturation (normally takes two days), females and males are easy to identify in that male D. suzukii have a pair of black spots on the tip of the wings. However newly emerged female and male flies are relatively harder to distinguish. In this case, the most easiest way is to look carefully at the tip of the abdomen. For female, we can see the serrated ovipositor composed of two symmetrical and sharp petals.
For male, we can see instead of two petals closing up like in female, two sclerotized, dark, “canine teeth” openning up, which are the genital claspers of male.
Note - “body size” is not a good indicator. - CO2 might impair the flies’ certain functions, so give fly two days to recover for future experiments.
